My Interest in Isolation and Identification of Escherichia coli from Urine Sample

The greenish shining colony in M-endo Agar distinguish E. coli from other Coliforms that help to separate E. coli from other bacteria present in stool and urine sample. Also, this method and media can be used to check contamination after antibacterial assay of E. coli. This method saved my time to identify by various sub-culture test.

Isolation and Identification of Salmonella spp. from Clinical/Food Sample

               The red colonies with black centers and media turn red (phenol red as indicator) in Xylose lysine deoxycholate (XLD) agar media distinguish Salmonella spp. from other coliforms that help to separate from other bacteria present in clinical blood/stool and food sample. Also, this method and media can be used to check contamination after antibacterial assay of Salmonella spp.
               Salmonellae can metabolize thiosulfate and produce hydrogen sulfide, hence black centers seen in colony but Shigellae can't and remains pink colony. Other coliforms are seen as yellow/orange colonies.
                Thus I separated Salmonella spp. very quickly from rest of the organisms.

We headed towards Koteswar from Balkhu, we were five talking about our honesty and innocuity. There was the conductor who had listened our expression. At destination, he asked for fare in greater rate, we discussed a while, but we gave the amount that he asked (greater than ususal).
Then one friend told that he had listened about our innocence, so he asked more fare and we could't resist.

The New Baby
Little Birkhe 's next door neighbors had a baby. Unfortunatly, the little baby was born with no ears. When they arrived home from the hospital, the parents invited Little Birkhe's family to come over and see their new baby.

Little Birkhe 's parents were very afraid that their son would have a wise crack to say about the baby so the dad had a long talk with little Birkhe before going to the neighbors.

He said "Now, son... that poor baby was born without any ears. I want you to be on your best behavior and not say one word about his ears or I am really going to spank you when we get back home."

"I promise not to mention his ears at all" said Little Birkhe.

At the neighbor’s home, Little Birkhe leaned over in the crib and touched the baby's hand. He looked at it's mother and said "Oh what a beautiful little baby".

The mother said "Thank you very much, Little Birkhe."

He then said, "this baby has perfect little hands and perfect little feet. Why... just look at his pretty little eyes.... Did his doctor say that he can see good?"

The Mother said "why, yes... his doctor said he has 20/20 vision.

Little Birkhe said "well, it is a darn good thing cause he sure couldn't wear glasses!!!

Involvement of protein kinase C- in DNA damage-induced apoptosis

Caspases are essential for the execution of cell death by apoptotic stimuli.1,2,3,4 The pathway of cell death varies depending on the cell type as well as the apoptotic stimuli. It is generally believed that binding of Fas ligand or tumor necrosis factor- (TNF) to their receptors causes activation of the initiator caspase-8 followed by the activation of a caspase cascade to execute cell death.2,5 In contrast, DNA damaging agents are known to induce release of mitochondrial Cytochrome c, which facilitates the interaction of apoptotic protease activating factor (Apaf-1) with procaspase-9 to initiate the activation of downstream effector caspases, such as caspases-3 or -7 to cause cell death.3 Both receptor-mediated and anticancer drug-induced apoptosis may, however, involve more than one pathway and there may be cross-talk between these two pathways.6,7,8

cis-Diamminedichloroplatinum(II) (cDDP or cisplatin) is one of the most important anticancer agents used for the treatment of solid tumors.9 Although the antitumor activity of cDDP is believed to be due to its interaction with chromosomal DNA, only a small fraction of cDDP actually interacts with DNA and inhibition of DNA replication cannot solely account for its biological activity.10 The efficacy of chemotherapeutic drugs not only depends on their ability to induce DNA damage but also on the cell's ability to detect and respond to DNA damage.11 cDDP, like other chemotherapeutic drugs, causes activation of caspases although the sequence of events that follow cDDP-induced DNA damage and lead to apoptosis remains to be unraveled.

We and others have shown that the PKC signal transduction pathway regulates cell death by cDDP.12,13,14,15,16,17 PKC is a family of 11 isozymes that are classified as the conventional PKCs (, I, II and ), novel PKCs (, , , and ), atypical PKCs ( and / ) and novel/atypical PKC.18 PKC is a substrate for caspase-3 and the catalytic fragment of PKC has been directly associated with apoptotic cell death.19,20 We have, however, demonstrated that downregulation of PKC that decreased the abundance of PKC catalytic fragment was associated with increased cellular sensitivity to cDDP.17 These results raise the possibility that PKC acts upstream of caspases to regulate cell death by cDDP. It is not known which PKC isozyme regulates activation of caspases and which step(s) of the cisplatin-induced cell death pathway is regulated by PKC.

Mitochondria play a pivotal role in the decision making process of a cell's life and death.21 It is believed that once Cytochrome c is released from mitochondria, cells are committed to die.22 An inability to induce release of Cytochrome c from mitochondria has been associated with cellular resistance to anticancer agents, including cDDP.23 In the present study, we have investigated how PKC regulates release of Cytochrome c and activation of caspases that emanate from mitochondria. Our results show that in HeLa cells, PKC was localized not only in the cytosol but also in the mitochondrial fraction and cDDP induced processing of both cytosolic and membrane-associated PKC. Rottlerin, a specific inhibitor of PKC, blocked cDDP-induced activation of caspases and proteolytic cleavage of PKC in both cytosolic and HM fractions but inhibited only late but not early release of Cytochrome c. Furthermore, inhibition of nPKC, but not of cPKCs, protected cells against cDDP-induced cell death. Taken together, these results demonstrate that PKC influences cDDP-induced cell death by acting at an early step of the mitochondrial cell death pathway that precedes activation of caspases.

Camphor

Description
The camphor tree is a dense broadleaved evergreen that is capable of growing 50-150 ft (15.2-45.7 m) tall and spreading twice that wide with a trunk up to 15 ft (4.6 m) in diameter, though the largest U.S. specimens are only half that size and those in the Caribbean are even smaller. The shiny foliage is made up of alternate 1-4 in (2.5-10.2 cm) oval leaves dangling from long petioles. Each leaf has three distinct yellowish veins. The outer margins of the leaves tend to be somewhat wavy and turn upward. The new foliage starts out a rusty burgundy color, but the leaves soon turn dark green on the upper sides and paler green underneath. New branches emerging from the shallowly fissured grayish brown trunk are smooth and green. Twigs are usually green, but may be tinged with red when young. The inconspicuous tiny cream colored flowers are borne in the spring on branching 3 in (7.6 cm) flower stalks. They are followed by large crops of fruit, comprised of round pea sized berries attached to the branchlets by cuplike little green cones. The berries first turn reddish, then ripen to black. Camphor tree can be readily identified by the distinctive odor of a crushed leaf.


This camphor tree shades a house in North Florida - a landscape use no longer considered appropriate for this invasive species.
Location
Cinnamomum camphora, the camphor tree, comes from China, Japan, Korea, Taiwan, and adjacent parts of East Asia, where it grows in mesic forests and on well-drained sites along streambanks. Camphor has become widely naturalized in Australia. In the United States, it is grown along the Gulf Coast and in California, and has escaped cultivation and become naturalized in many areas.

Culture
Camphor prefers fertile sandy soil. It will tolerate a pH anywhere in the range of 4.3 to 8. The roots are very sensitive to disturbance. They may extend far from the trunk of the tree, and can readily be identified by their characteristic odor.
Light: Camphor will grow in full sun or partial shade.
Moisture: Camphor tree does not do well in wet soils. Established trees are tolerant of drought.
Hardiness: USDA Zones 8 - 10. Hardened off camphor trees can survive freezes down to 10-15ºF (-12 - -9ºC), but new growth will suffer freeze burn when the temperature drops below 32ºF (0ºC) and branches will die back from temperatures in the low twenties.
Propagation: Camphor seed does not remain viable for long and should be planted in the greenhouse as soon as it ripens. Remove the fruit pulp first. At 68ºF (20ºC), germination will take 1-6 months. Cuttings of semiripe side shoots can be rooted in a warm humid place in midsummer. Pieces 2-3 in (5.1-7.6 cm) long with a heel work best.

Usage
Camphor is widely planted as a shade tree, screen, or windbreak. In China and Japan, it is grown commercially for its medicinal oil. Camphor oil has a strong penetrating fragrance, a pungent bitter flavor, and feels cool on the skin like menthol, though it also has irritating qualities as well as a numbing effect. Camphor has been used to treat ailments ranging from parasitic infections to toothaches. Scientific evidence has confirmed that chemicals in the plant have value in antiseptics and medications for treating diarrhea, inflammation, itching, and nervous conditions. Camphor wood is prized for its attractive red and yellow striping, amenability to woodworking, and insect repelling properties. It is light to medium in weight and soft to medium in hardness. Wood from the camphor tree is not especially strong, but it takes polishing well. It is commonly used for chests, closets, coffins, instruments, and sculptures. Camphor veneer is used in fine cabinetry. Camphor is also used in perfumes.

Features
This is a sturdy storm resistant tree which makes a good windbreak. Since it is hard to burn, it should also be valuable as a shade tree in areas that are prone to wildfires. Unfortunately, these desirable traits are offset by the tree's invasiveness and damaging effects on wildlife and natural communities. This fine tree should be grown and appreciated in its native range, but not planted in other regions where species and ecosystems have not adapted to its aggressiveness and toxicity. Camphor tree should not be grown in the United States.

WARNING
Camphor in large doses is toxic to humans. It stimulates the central nervous system and may affect respiration or cause convulsions. In Chinese medicine, camphor is forbidden for pregnant women and those with a deficiency of vital energy or yin. Camphor is a prolific seed producer that apparently does not have serious predators or diseases outside its native range. Seedlings and root sprouts are abundant near mature trees, but individual trees pop up far from seed sources. In Florida, camphor trees appear in undisturbed mesic hardwood forests, upland pine woods, and scrubs, as well as in the vacant lots and fencerows where it is more commonly observed. The Plant Conservation Alliance lists this species as an Alien Invader and it is listed as a Category I invasive exotic species by the Florida Exotic Pest Plant Council, which means that it is known to be "invading and disrupting native plant communities in Florida

U.S. scientists are using a new form of scanning microscopy to simultaneously study physical and electronic profiles of metal nanostructures.

Researchers at the National Institute of Standards and Technology and University of Colorado-Boulder say the new instrument — the scanning photoionization microscope — is expected to be particularly useful for analyzing the make-up and properties of nanoscale electronics and nanoparticles.

The scientists said the instrument combines the high spatial resolution of optical microscopy with the high sensitivity to subtle electrical activity made possible by detecting the low-energy electrons emitted by a material as it is illuminated with laser pulses.

The technique potentially could be used to make pictures of both electronic and physical patterns in devices such as nanostructured transistors or electrode sensors, or to identify chemicals or even elements in such structures.

“You make images by virtue of how readily electrons are photoejected from a material,” said NIST fellow David Nesbitt, leader of the research group. “The method is in its infancy, but nevertheless it really does have the power to provide a new set of eyes for looking at nanostructured metals and semiconductors.”

The instrument is described in the journal Chemical Physics.

A Week Without Contact of Friends

A Week of few Word Uttered
       Everyday we used to meet at a certain point that would be almost at equal distant for all of us from our room. We used to debate Microbiology, and I was loquacious. If we could not meet, we used to phone one another. But last week became different to me. I did not meet anyone and surprisingly NO phone. I could utter just 2-3 sentences/day, a thrift week.
       I had spent 3 days in IT unit central library where I had spoken only the word, “Namaskar Lal sir.” One day I had planned to meet Khagendra Koirala who had invited me through SMS. But on the road I lured and stuck on cyber café, meanwhile I got an SMS again that he was not in his room, so I continued for five hours in Nayabazaar. Rest of the days I spent in the apartment.
       I was not angry with any of the friends and neither disappointed nor diseased. It was fortuitous. I showed the insanity to study Visual Basic 6.0, VBScript, and web page designing, which I had started to learn many years ago. I wanted a little bit advance study. Fortunately, in the halcyon environment I could study in deep and could generate some files and completed some basic tasks. I could make some amusing web pages as well as some calculating software (that could be useful in our practical class) in Visual Basic. Now I laugh at me why I was spending so many times in computer with such craziness. My profession is not for the computer because my degree in biology.

Gossip, Tea and Hidden Reality
It was the month of January, the weather condition was cool due to which several living structure was facing a harsh momentum. The appearance of massive dews can be seen by every eye’s at the first period of shining moment of the day.
With running time of morning,Ramesh ‘a social worker’ get up from the bed. After having mouth and teeth washed and brushed, he takes a cup of tea prepared by his native, as he was living their in the position of social worker since fifteen day before so he was quiet familiar with common existing system.
He started to walk towards the town which was a kilometer away from the place where he uses to stay. The town was the place to have a gossip among the people of several society, casts creeds and religion. For them morning was the right moment to have a first meet. No any topic used to remain constant for gossip. With the changing moment, the topic of gossip used to change day by day. But usually they use to talk about the social, political and economical aspect. It doesn’t mean they do not talk about other mater; they use to talk about other mater but it seems to be in the rear case even they use to talk about various absurdities.
After having a walk of fifteen minute Ramesh reach the town and started to find out his destination, where he could find the group of people whom he is feted for to have a gossip by the medium of tea ‘sitting at the corner of the hotel. Finally he found the symbol that he is searching.

Hello! Ramesh, how are you? Let’s have a hand check one of his friend shouted from the corner side of hotel which is located at his left hand side. Same process of hand check repeated one among another among the people who are present their and who are well known to him. They odors one more cup of tea and start to have a gossip that they usually use to have.
They all are professional personality, so usually they use to have a talk about several professions. It can be said that there is a collection of people who are engage in several sector. Among them some are social worker, some are businessman, some are politician and some are teacher. But there is still the majority of social worker. So due to its majority, mostly the topic used to be about social mater. Same process repeated hat day also.
While they were having a gossip, the men who seem very old came on the hotel and ask for the cup of tea to he owner. His dress seems to be very dirty which was symbolizing that he was from a very poor family.
Ok! I will provide you after a while ‘the owner replied with hesitative sound.
Among the massive presentation the old man sit separately with gloomy face.
The hotel owner was making the tea for people who came after the old man but he was not showing the sign to provide the cup of tea to the sick old man.
With curious nature Ramesh along with his friend are watching the event that is being going on which move their mater of concentration towards next direction
With the shivery mode because of cold, the old man was waiting for the tea at the next corner of the hotel with solitary nature.
Pointing towards the owner Ramesh shout, why are you not providing the cup of tea to this old man since he is waiting for a long time? Sir he was ‘dalit’ the person of lower cast, owner replied.
The reply seem very hypocritical to Ramesh. He found their still remaining the sign of extreme castism.
The hotel personal bring the cup of tea towards the old man and odors "take and clean the glass" with a rude manner. Ramesh rub his eyes to notify whether he is in dream or not. Finally he found that he is not in the dream.
Is the system of cleaning he glass is for everyone? If so why the before coming personality went without cleaning the glass? He asks to the owner.
With calm manner the owner replied –‘sir, they are not a dalit, i.e. they are the people of higher class but he is dalit so he have to clean the glass.
If we clean his glass what the society will say! Sir? He adds.
Ramesh found Thick absurdity with thin reality in his saying.

It is the system having origin point on past and is still going on. But now we have to stop its continuity. One of his friends sitting with him tells.
Of-course ‘it is old contradiction which is against of true humanism, so we have to pull out its root.’ Ramesh add and move towards the old man.
Grandfather, what is your name?
Ram bahadur sharki, he replied.
What is your profession, grandfather? Ramesh again asked.
At past we use to make the shoes but now a days situation change, instead of ordering us to make the shoes people started to go for a town to have a shoe. So now I use to work as a house builder, but I don’t know up to when it last for? I am hearing, in big towns and cities engineers have started to construct the house. He replied.
Ramesh found the mass of truth in his innocent eyes. The discussion last for a hour.
Ok! Sir, I am getting too let as I have to go for a work. The old man said and started to move towards the place where he could clean the glass.
Ramesh came towards the old man and snatched the cup from old mans hand. With the glass he go in front of the owner who was in the counter and said, if you want to give continuity to your business start o treat equally among all people.
The owners face turns red. He odors his personal to carry the cup.
Ramesh pay the money for old mans tea. And their physical structure became temporarily departed.

Moment of Dusk
It was a moment of dusk when I was returning ' from the nature. Nothing was there to hinder me up and was shocked. I was in my own way and recalling all the moment or event that occurred.
Several symbol was their which was applied that brought the mass of fuliginous and non fuliginous resultant. The resultant was their ‘the solution of morality and non morality, sociality and non sociality and also there existing a mixture of symbiotic and parasitic structure.’ Several blossoms was their, who became the target point and the result published where the sprit of the blossoms was make demised. Though the heart weeps a tear, the collogues remained quiet and was locked in the cage within the limited area providing the four sided agent for the purpose to concentrate towards the vibration of their physical structure. "The social evacuees" was the title rewarded to the up coming offspring even they were respiring in the womb of female vegetation.
I used to visit their physics. I used to find the physical gloomy, which used to elongate my journey and extended up to their psychology. Because I was unable to create the satisfaction with its limited edition. When I reached there, I found self in the next side of round sphere and the condition that I found was full of more and much fear that was ever and even expected by my sorts of social structure before.
The physical nature was same as statues which was silent, which is silent and which will be silent forever. And, no symbol was remaining there for recovery which was incurable fever.
When I was walking on the world of their wants, desiration and psychological needs one of their candidates arrived in front of me. With gloomy nature, she requested me to carry her up in the world of pigeon from that of vulture. " Manage some space and take us away, we have no more toleration power to create the resistivity force for resisting the torture provided by the destructive diabolic spirit try to let us to escape from the world of cactus and provide to move towards that of orchids" was the prey of their soul. But it was me who can do no ever than providing a consolation which was the weakness having origin point on me.
It began to make me disturbed. Each and every spark of my day and night started to symbolized me a agent which have a way but no destination, a moon which have to depend upon other to maintain its position.
It made me unable to have a stand within the same position. So, I refused to be limited within the moon and started to step towards the sun Also I left to be the product and started to follow pre required procedures to be the producer.
It was my first experiment, which brought several, ups and down in the life system of several social being, it was the experiment which carry away the physical existence of several spirit and also the attempt which successfully break the ice and replace the several ex- existence which have been threatened.
It was a time when I have just started to step towards the sun, one devil came in front of me and create a block in my way. He applied as many procedures as he can, to make me limit within the moon. However, I strongly rejected it, as I never wanted to do it because just a day before I was waked and never wanted to be slept again.
When I did it, mass of agony was observed in his eyes, which were symbolizing that he was going to swallow me alive. ‘carrying the agony in both nature he came towards me and shackled my hand due to which it became unable to write, my happiness was threatened which include black and remove the symbol of bright as only the physical nature was free and all the psychology was imprisoned. So once again the blossoms expectation remained unfulfilled, again they got compiled to adjust evil spirit. The future that they have started to see was re-eroded. As they began to feel that earth stop to rotate. However, the situation does not remain stable. The next blossoms came more revolutionary then ex-blossoms. They started to pick up the voice of liberation, as a result diabolic spirit became unable to face and flew away which brought sign of freedom and each and every chain psychology were set free, each and every soul gained a right to live under the canopy of owns aspiration as well as desiration in a same momentum , I was also awarded a freedom.
Today all the hunted wants are going to be overcome and I am capable to go in nature and return from there without any hesitation. There is no any agent to create a block in my way but in future, there may be the creation of next block. In against of such block, I fought a past and if needed I will fight in future. As the group of upcoming blossoms in my sight to fight against such non moral creation.

I shook hand with sir
When our department was locked, I shook hand with a sir who was standing in front of channel gate (door), thinking that he was our one batch senior dai and said, "K chha dai? Aaram hununcha?" giving my hand.

Forgot
One of our sir told me to deliver that he would not coming tomorrow to teach us and would come on Tuesday. "Tuesday?" ,I said. "Yes don’t forget to deliver",sir emphasized. "Ok sir". After half an hour when friends met, I said, "sir is not coming tomorrow, he will come on Sunday." (How quickly I forgot).

One of our, friend expressed his feeling while entering for tea on a house whose was made up of wood and mud and was squabble. He said, "Oh! like my house, I am feeling I am in my house". Then another friend replied promptly, pointing towards house owner who was cooking tea for us, "He may be your father".

Dog thought
In a picnic spot a dog staring a friend for a long time. The friend was then astonished and told, "Why is it looking at me so long time?" then other friend said, "Perhaps it is thinking about his big brother?"

I realized mistake.

I got in a microbus from Kalanki for Koteswar toward Sanjeev's room and sat in front sit (not in cabinet). In Balkhu a girl (may be married woman) and a middle aged man entered the microbus and sat in the same sit. The girl was in between me and the man I was in corner toward the window.
I looked outward (I prefer in journey to view seen) and had never bent toward the girl and door after she entered. In Satdobato wari she got off from the microbus. The helper then asked for fare all of us. While giving fare I bent toward the helper (in door side) and I found a fifty rupee note in the sit. Conforming that money was not mine I asked the middle aged man if that belongs to him because he has just taken out money from his pocket to pay fare. At the same time he was giving fare telling he will for Tinkune. After taking money he promptly got off from the microbus in Satdobato pari. Then I realized my mistake.

Today I had gone to observe can infotech.
I think this is only for business view where latest technology is advertised to lure people and grab their money. It would be very good to exibit how to use them for revenue generation or to simplify life or how we nepali also can make such device.
It would be better to use Nepali experties to export software in foreign countries or to use them to make software used in Nepal at least to displace foreign technology.
